IL-12 drives the differentiation of human T follicular regulatory cells

چکیده
Abstract T follicular regulatory (Tfr) cells can counteract the B cell-helper activity of helper (Tfh) and hinder production antibodies against self-antigens or allergens. A mechanistic understanding cues initiating differentiation (Treg) into Tfr cells, which is instrumental for therapeutic manipulation diseases associated with a cell imbalance, still missing. Despite their opposed roles, Tfh appears to be controlled by partially overlapping signals, including TCR stimulation costimulatory molecules. However, it unknown if cytokines that have been shown control biology human IL-12 activin A, influence cells. To address this question, we evaluated impact these two on in vitro Herein, report role driving, activated Treg induction molecules belong program, CXCR5, PD-1, BCL6 ICOS meanwhile preserving function. DIFFERENTIATION OF HUMAN OVARIAN FOLLICULAR GRANULOSA CELLS INTO KERATINOCYTES
Background & Aims: Stem cells are undifferentiated cells and are found in different tissues. These cells have capacity of self-renewal and differentiation into other lineages. Granulosa cells (GCs) are the multipotent stem cells. In the present research we evaluated the differentiation potential of GCs into keratinocytes. Material & Methods: GCs were cultured after enzymatic isolation from ova...
متن کاملT follicular helper cells, T follicular regulatory cells and autoimmunity.
CD4(+)T follicular helper (Tfh) cells are recognized as a distinct T-cell subset, which provides help for germinal center (GC) formation, B-cell development and affinity maturation, and immunoglobulin class switching, as an indispensable part of adaptive immunity. Tfh cell differentiation depends on various factors including cell-surface molecule interactions, extracellular cytokines and multip...
متن کاملEnforced Expression of CXCR5 Drives T Follicular Regulatory-Like Features in Foxp3+ T Cells
CXCR5+ T follicular helper (Tfh) cells are associated with aberrant autoantibody production in patients with antibody-mediated autoimmune diseases including lupus. Follicular regulatory T (Tfr) cells expressing CXCR5 and Bcl6 have been recently identified as a specialized subset of Foxp3+ regulatory T (Treg) cells that control germinal center reactions. In this study, we show that retroviral tr...
متن کاملIFN-alpha and IL-10 induce the differentiation of human type 1 T regulatory cells.
CD4(+) T regulatory type 1 (Tr1) cells suppress Ag-specific immune responses in vitro and in vivo. Although IL-10 is critical for the differentiation of Tr1 cells, the effects of other cytokines on differentiation of naive T cells into Tr1 cells have not been investigated. Here we demonstrate that endogenous or exogenous IL-10 in combination with IFN-alpha, but not TGF-beta, induces naive CD4(+...
متن کاملIL-17+Foxp3+ T cells: an intermediate differentiation stage between Th17 cells and regulatory T cells.
Foxp3(+) Tregs have been known as a major regulator of immune homeostasis through their immunosuppressive function. Th17 lineage is a CD4(+) T cell subset that exerts its function by secreting proinflammatory cytokines and protecting host against microbial infections. The altered ratio between Foxp3(+) Tregs and Th17 cells plays an important role in the pathogenesis of immune-related diseases. ...
